I've got 35 hours so far, which isn't a lot for a game like this, but it's sure as shit longer than I spend on most games, and as you play, the differences between this and Civ5 just get more and more pronounced:
1. Strategic resources are different. No more "get salt, win the game".
2. The orbital layer is completely new.
3. Aliens are way more interesting/involved than barbarians. Christ, you can turn aliens into allies.
4. Tech Web vs. Tech Tree, not even close.
5. Unit upgrades via affinity vs. tech is COMPLETELY different, and way more fun, if you ask me. I like rushing Supremacy to get CNDRs first. Then you die to my robot army.
6. Other stuff I'm forgetting.
So yeah, it's still food, hammers and beakers, but after a while the differences are way more noticeable, and in this author's opinion, are generally improvements.
